The RENEWAL-NAD+ trial

Summary

What the study found

RENEWAL-NAD+ was a double-blind, randomized, placebo-controlled trial in healthy adults aged 45–75. Sixty participants were randomized and fifty were included in the primary analysis. Participants took oral LNAD+ or placebo for five days.

Intracellular NAD Rose 53% versus placebo at Day 6, already detectable at Day 4 (p = 5.48×10⁻¹⁴)
Circulating plasma NAD Unchanged (p = 0.60) — a compartment-selective response
NAD metabolites MeNAM and 2PY rose sharply, indicating active metabolic processing
Safety Well tolerated. One mild adverse event, resolved within 24 hours. No serious adverse events.

Why compartment selectivity matters

NAD+ is consumed inside cells. An increase measured in the bloodstream does not show that the molecule reached where it is needed. Our trial found the opposite pattern to precursor and intravenous approaches: NAD rose inside cells while plasma levels held steady.

Research ongoing

The trial included multi-omic endpoints specified in advance. These analyses are hypothesis-generating and are shaping the design of our next study. Results will be reported in a forthcoming publication.

RENEWAL-NAD+ dosed participants for five days and was not designed to assess long-term effects. It enrolled healthy adults, so the findings do not establish benefit in any disease population.
